Webb1 jan. 2016 · Polyclonal rabbit antibodies (rIgGs) were purified from 2 ml of commercial rabbit serum by affinity chromatography on a 5 ml HiTrap protein A column (GE Life Science) according to the manufacturer's instructions. Binding was performed in 0.02 M sodium phosphate (pH 7), and rabbit IgG was eluted with 0.1 M citric acid (pH 3). WebbProtein A ligand is recommended specifically for polyclonal IgG from rabbit, pig, dog, or cat serum. It binds the heavy chain constant region (Fc) of IgG (CH2-CH3 region). Native Protein A is purified from Staphylococcus aureus (46.7 kDa; 4 IgG-binding sites).
